I don't think it will work as the .45ACP headspaces on the case mouth, and the .45LC on the case head. If you tried loading the .45ACP in the .45LC R&D cylinder, the ACP rounds will drop some distance into the chamber. You'll need something to poke them out. Perhaps you could try a Kirst Cartridge Konverter in .45ACP. I converted one of my Pietta 58's to .44Remington, my other Pietta '58 uses Kirst's .44Remington cylinder with their old model plain back plate. No ejecting rod needed, I swap cylinders ala "Pale Rider". Lots possibilities, too little money... Good shootin'!

I actually had a cylinder made for my Pietta New Army in .45acp. I only shoot 21 grains of GOEX FFFg and a 155 or 200 grain bullet. I was planning to have a .44Colt cylinder made for the original heeled bullet but the Smith lost interest. His firing pins weren't as successful as I'd hoped, so I didn't push it.
I think most smiths would shy away for fear of some doofus putting IPSC "major" ammo through it.

GCR; You are right on! Much the best solution if you have an R&D cylinder in .45Colt.
I wanted a .44 Colt, so the smith cut a star at the rear of the cylinder body for indexing rather than the pin used by R&D. It allowed a .483 rim diameter but not enough for the .45 Colt rim. As I have a lifetime supply of .45ACP brass, I asked him to make a separate cylinder body for that round. The .44 Colt body never came off the mill!

Folks, using the Cowboy .45 Special cases and DD's Big Lube bullets, these conversions are happy, happy. I heard from Walt Kirst himself that the C45S is the best choice for these guns, as they are easier on the GUNS over time, and will make ya happier longer.

As far as shooting .44Remington or .44Colt heeled loads, Gad's Custom Cartridges loads for these two cartridges and they work well in my two Pietta Remington NMAs. The former load uses a 248 grain heeled base (outside lubed) bullet pushed by your choice of smokeless, Goex black powder, Pyrodex P or Triple7 FFFg. I prefer the last powder as my guns clean up pretty quickly with Ballistol "moose milk" (10:1 mixture of water to Ballistol) and I've had no problems with rust. The link to his website is below.
